Mutation is the random process with regards of to the structure and size of the gene. The possibility of damage or abolish the function due to mutation of gene is always there. The mutation in the gene can only affect the product (RNA or protein) coded by mutant copy of the gene or may not affect the product code by any other allele. Two mutant fails to complement each other or produce wild type phenotype if they are present in Trans configuration in a heterozygote (or they are part of same gene)

The null mutation refers to the complete loss of gene function, this is because of deletion of gene. If the gene is essential a null mutation is lethal when in homozygous.

The other mutant is showing dominant negativity means alteration of gene product that act antagonistic to wild type mutant. Such event causes the alteration of overall molecular function will be dominant.
