Student A calls you over and explains that after running his column and showing you the TLC of his first 5 fractions, he collected the first spot, removed the solvent and took a melting point. He is worried something is wrong because he got 38°C and not 133°C How would you advise him? HW 2. Student B calls you over and explains that after running her column and collecting 25 fractions, only one spot has come off! What would you advise her to do? HW1. Don't worry, is probably that you didn't remove solvent completely. other option is to evaluate other TLC's spots.. the worst option is your sample is contaminated.
HW2. no problem, you did it.. if you want improve the elution, you can to change the polarity of the movil phase, changing the solvent or solvents proportions.
