plz help me answer question

Medgar Evers College Bio 302-Genetics- Problem Set 5 Prof. Small 11.25.13 be a) Suppose you label dividing cells wi 32 In which DNA molecules will the radioactivity incorporated if you were to look during metaphase? b) Suppose you are examining the above cells after replication. In which DNA molecules will the radioactivity be present? 2) In one picture, draw a schematic indicating how DNA is synthesized bi-directionally. The schematic should indicate which are the lagging strands, which are the leading strands, wher RNA primers are, and the 3 and 5 orientation of the strands Indicate the Okazaki fragment the figure. b) Why does the lagging strand have Okazaki fragments while the leading strand does no 3) Suppose you had a bacterial mutant that was defective for topoisomerase, one that was defective for primase, and one that was defective for DNA polymerase. Your student mis- labelled the three cultures. How could you differentiate among the three mutants 4) Describe two ways in which prokaryotic and eukaryotic replication differs

Explanation / Answer

Answer 1: A) Cell cycle consists of INTERPHASE and M (mitotic) phase. Interphase is the period of cell growth and DNA synthesis. M phase is the dividing phase and consists of prophase, metaphase, anaphase and telophase. Since DNA synthesis occurs during interphase (specifically S-phase) so if cells are labeled at metaphase , 32-P won't get incorporated into the DNA. Thus, there will be no radioactivity in DNA. B) However if we observe after one round of replication then 32-p radioactivity will be observed in the newly synthesised strand (daughter strand).
