This is a TLC spinach pigments lab. This question is important to me so pls only answer if you're sure 4) Indicate what might have happened in each one of the following cases that are observed during the TLC analysis of a spinach extract. In other words, write down the reason(s) as to what could have caused the following problematic observations: a) A wide diameter capillary tube instead of one with a fine diameter was used to spotting the sample on the "spotting" line of the TLC plate. b) The component spots during development are not moving up the plate as separated spots but as a tailing band or a streak. c) The separated spots during development are not moving vertically or straight up the plate but in a slanting manner

Explanation / Answer

(a) Too thin capillary tube may prevent the uptake of sufficient amount of sample.

(b) Streak or tailing could be because of 2 reasons:

1. If the sample is too concentrated to be handled by solvent while moving up.

2. If the sample is ionic or highly polar and bind strongly to solid phase.

(c) It is because the bottom of the plate may not be parallel with the solvent level but at an angle.
