Question

1) Ethanol is supplied in a 90% solution. You need 750 ml of 75% ethanol for your experiment. How would you make this solution? 2) you need to treat cells with aresnite such that it has a final concentration of 50 uM once it has been added to the cells. You need to treat 10 mL of cells, and you don't want your treatment volume to be more than 10 uL per 1 mL of cells. What conversation should you make your stock solution? 3) TAE agarose gel running buffer is sold at 50X concentration you will need 12L of this solution at a concentration of 1X for ready use in the lab. How would you make this solution? I need the right and full solutions to these questions asasp 1) Ethanol is supplied in a 90% solution. You need 750 ml of 75% ethanol for your experiment. Explanation / Answer

We can use rule of S1V1=V2S2;

where S1= strength of solution 1, V1 = volume of solution 1, S2=strength of solution 2 and V2= volume of solution 2

1) say x mL of 90% = 750 mL of 70% , thus X*90 = 750*70 ; X= 583.33mL

For making 750 ml of 75% ethanol, we should take 583.33mL of 90% stock solution and rest should be fill up with solvent generally water.

2) volume of arsenite solution should not to be more than 10 uL per 1 mL of cells. thus for 10mL of cells, maximum limit is 100uL .

Thus 100uL of x concentration should be equal to 50 uM of 10 mL ;

100 uL * x = 50 uM *10 mL ; 100*10-6 * xL = 50*10-6*10*10-3 ML ; x= 50*10-6*10*10-3 M/100*10-6 = 5*10-3 M

So concentration of stock solution should be 0.005M.

3) 50X * Y mL = 12 L*1X ; Y= 12/50 = 0.24 L

thus by taking 0.24 L of 50X contration solutionof TAE agarose gel into the volumetric flask and the diluting it to 12L we can get our required solution.
