Question
Can you explain in a paragraph what does this figure mean and explain the data results. Please include biochemical terms in the explanation. Thank you 3:21 Carbohydrate Discussion Sore Joints A contributing factor to the development of arthritis is the inappropriate proteolytic destruction aggrecan component of cartilage by the proteolytic enzyme aggrecanase. The immune system signal molecule interleukin 1 (IL-1) activates aggrecanase; in fact, IL-2 blockers are sometimes used to treat arthritis. Studies were undertaken to determine whether inhibitors of aggrecanase can counteract the effects of IL-1. Pieces of cartilage were incubated in media with various additions and the amount of aggrecan destruction was measured as a function of time of the Bovine nasal cartilage was incubated in control media (squares), IL-1 (circles), or IL-1 plus the selective aggrecanase inhibitor, ST154 (triangles), and media were removed and replaced with fresh treatments every 2-3 days. Media were analyzed for agg degradation products and cumulative glycosaminoglycan (GAG) released per mg wet weight cartilage are plotted versus time of incubation using an IL-1 stimulus of 5 ng/ml (A) or 2 ng/ml (B) recan Start Thread
Explanation / Answer
A) The rationale for this assay is that glycosaminoglycan is not usually released during the normal conditions. But during aggrecan degradation we see that there is release of GAG. That is why release of GAG is measured.
B) GAG release doesn not indicate aggrecan degradation because it is released in excess only initial time later the release of GAG is decreased.
C) Every experiment is controlled because it provides a baseline to which the results are compared. It helps us to determine the change that occurs in the test sample.
D) The effect of IL-1 is visible during the immune response because all the immune responses are mainly regulated by IL-1.
