Can anyone elxplain to me how in the figure below the loss of ion channel activity (TRPV3) is lost?

TRPV3 agonist cocktail (200 mM2-APB + 500 mMCarvacrol) induced large increases of [Ca2+]i in primary cultured keratinocytes isolated fromWT(V3+/+) but not V3KO(V3/) mice. Whereas more than 80%of WT keratinocytes responded strongly to the V3 agonist cocktail, negligible responses were observed for V3KO keratinocytes. Positive controls:60%–80% keratinocytes from both genotypes (WT and V3 KO) responded to 4a-PDD (3 mM; agonist of TRPV4). All cells responded to ionomycin (1 mM).
Explanation / Answer
Application of TRPV3 agoinst cocktail (200 M 2-APB + 500 M Carvarcol) induced large increase of Ca2+ ion in primary cultured keratinocytes isolated from wild type (WT or V3+/+) but not V3 KO (V3-/-) because the removal of extrernal Ca2+ abolish most of the agonist, which effects the Ca2+ responses. Hence, there is no singnificant increase in Ca2+ in primary keratinocytes V3 KO. Keratinocyte TRPV3 sensitivity depends on the Ca2+ -permeant channels, which forms a signaling complex with TGF-/EGFR.
