Question

1) the following experiment was conducted to observe theeffect of temperature on the rate of enzyme activity.      - 10 mL of a starch solution wasadded to each of the five lettered test tubes      - Each test tube was placed in adifferent water bath as shown below       - An equal amount ofsalivary amylas was added to test tubes W X Y and Z       - A sample was taken fromeach test tube every minute and tested with IKI, an indicator thatturns from yellow to black when mixed with starch test tube temp of water bath 1 min 2min 3min 4min 5min V 20 black black black black black W 0 black black black yellow yellow X 20 black black yellow yellow yellow Y 40 black yellow yellow yellow yellow Z 60 black black black black yellow A) What is teh purpose of test tubeV?                     1 mark B) Explain the results of theexperiment                     3 marks 1) the following experiment was conducted to observe theeffect of temperature on the rate of enzyme activity. 1 mark B) Explain the results of theexperiment                     3 marks test tube temp of water bath 1 min 2min 3min 4min 5min V 20 black black black black black W 0 black black black yellow yellow X 20 black black yellow yellow yellow Y 40 black yellow yellow yellow yellow Z 60 black black black black yellow

Explanation / Answer

A) Test tube V serves as a control group. This is to make sure thatthere is no other variables except temperature manipulate theresult. B) In general, as temperature of bath increases, less time is requiredfor amylase to digest starch completely. This is because increasedtemperature provide more kinetic energy for the substrate andenzyme to collide so that reaction happens more readily. However, at temperature exceeds 40 degree celcius (60 degreecelcius as seen in the result), more time is needed for amylase todigest starch completely. This is because at very high temeprature, the three dimensional structure of amylase will change. As aresult, it lose its catalytic function.
