If both mice are of same species then they will interbreed. So we need check if both river side mice interbreed between each other. If their offspring comes with the same characteristic then they ate of same species.
All morphological anatomical characteristic should be noted before starting the experiment. Two mice from both places should be kept in same space and observe their food habits and reproductive behavior. If they can not produce any offspring then they are different species.
