Question
You are interested in studying lung cancer. You hypothesize that lung tumor growth is due to deletion of a gene required to stop the cells from dividing uncontrollably. You have both wild type and tumor lung tissue. Which of the following experimental approaches would be a definitive test of your hypothesis? EXPLAIN
a) Western blot comparing wild type and tumor cells.
b) Southern blot comparing wild type and tumor cells.
c) Northern blot comparing wild type and tumor cells.
d) PCR using genomic DNA prepared from wild type and tumor cells.
Explanation / Answer
To study lung cancer the best hypothesize method is using Option d that is PCR using genomic DNA prepared from wild-type and tumor cells as this diseases is related to gene dysfunction which can be the best study through genome study by which we can find out difference in normal gene and tumor gene as compared to other method given out it's quite simple and fast also as other blotting techniques requires much more time with more precise setup knowledge.
Moreover, PCR gives us more precise knowledge about gene defect as for western blot we required protein for southern blot we required ultimately DNA only and for Northern blot we will require an RNA which is not possible in this case.
As we are comparing two type of sample so PCR based approach allows us to compare two sample gene data more accurately.
