When culturing Chiamydomonas, ampicillin (amp) is added to cultures at 50 ug/mL to prevent bacterial growth. You are splitting your culture and maintaining your new culture at 50 mL. You look in the freezer and realize that there are no amp stocks available for your use but you know where the amp powder is kept, so you decide to make fresh stock. a. What is a reasonable concentration for this stock? Why? (you can choose to make any concentration of stock, it doesn't have to be what we use in class, but why would it make sense to make that concentration?) b. Show your calculations for your stock. c. How much of your stock must you add to your culture?Explanation / Answer
A) Reasonable concertation of Ampicillin stock solution will be 100 mg/mL. We make this concentration as serial dilution is easy with that.
B) Calculations for stock: Dissolve 1 g of sodium ampicillin in sufficient H2O to make a final volume of 10 mL.
C) Amount of stock to be add in culture: For bacteria cultures, the stock solution is to be diluted into the LB-Agar at a 1/2000e dilution (50µg/mL = final concentration).
