3: Complementation test determines if the two mutants are of the same gene or belong to the different ones. A gene regulate the production of a particular active enzyme. Its mutant allele regulates the production of the inactive form of that enzyme i.e. mutant phenotype.
Cis heterozygote has wild type alleles on one of the two homologous chromosomes. Presence of wild type allele ensure the production of active enzyme and there is expression of wild type phenotype. Expression of mutant phenotype by trans heterozygote shows that the two mutants belong to the same gene since it will produce inactive enzyme and mutant phenotype will be expressed.
