Cyclin E is an important regulator of the cell cycle. Along with cyclin dependent kinase 2, it is needed for the transition of G1 to S phase in the cell cycle. This initiates the process of DNA replication. The concentration of cyclin E increases drastically in the middle of the G1 phase. The concentration of cyclin E will be highest between the two phases of G1 phase and S phase. The concentration of cyclin becomes 0 in the middle of the S phase. The cyclin E is then degraded.
The gene FBXW7 encodes F-box proteins which tag cyclin E for ubiquitination and degradation. This gene undergoes loss of function mutation.
Cyclin E overexpression occurs in most of the tumors. Gene amplification is responsible for the overexpression of cyclin E.
