Explanation / Answer

UTI tests are used to diagnose the urinary tract infection. The causative organisms includes gram -ve bacilli (E.coli, Pseudomonas, Proteus etc.), gram +ve bacilli (Staphyllococcus, Enterococci), Miscellaneous (M.tuberculosis,Citobacter, Salmonella) and Fungus (Candida albicans). For the presumptive diagnosis of the bacteria we need to culture the bacteria by using different media like, Trypic soy Agar (TSA), MAC Conkey Agar, Mannitol Salt Agar (MAC) etc.

We can discuss the results of the above microbes culture at different media.

TSA Plate: This is used for the cultivation of non-fastidious bacteria,which are able to grow without special nutritional suppliments.

Results: E.coli,Staphylococcus aureus and Proteus Vulgaris are non-fastidious bacterias, so it can grow in TSA Plate gives a positive results.

MAC Plate:It is selective and differential medium. Most gram -ve bacteria will grow. But gram +ve bacteria are inhibited by the presence of bile salts and crystal violet inhibotors in the medium.

Results: E.coli and Proteus are gram -ve bacteria, so it acan grow gives a positive result whrereas Staphylococcus is a gram-ve bacteria can not grow in the medium.

MSA Plate: It is also both selective and differential medium, selective for Staphylococcus aureus. High salt concentration inhibits most bacterium.

Result: Only Staphylococcus aureus can survive give a positive result.

We can summarize like this;(+ indicate positive; - indivates negative). These results gives a presumtive diagnosis of the bacteria and shows the possible UTI for E.coli, S.aureus and P.vulgaris

Sugar fermentation for MAC and MSA:

MAC differentiate lactose fermenters from non-lactose fermenters. E. coli: strong lactose fermentation resulting in bright pink halo (bile ppt) and pink growth. P. vulgaris can grow but not ferment lactose.

MSA differentiate between staphylococcus sp. by sugar mannitol fermentation. Mannitol fermentation produce acids which change the medium pH. Peach colour indicates no fermentation and bright yellow colour indicates mannitol fermentation.
