We can isolate puc 19 palsimd contained bacteria by grow them in lactose and ampicillin added medium. Lactose is added because the plasemid has lacZ operon and only this type of bacteria can consume lactose sugar as their energy source. The second is antibiotic ampicillin which is present in plasmid as APR gene. If the bacteria have puc 19, the bacteria will survive in ampicillin condition.
