Question

3 3: Growth Kinetics You run a series of experiments to determine the relationship between substrate concentration (S. in mg rbCODL) and the specific rate units given bacterium. When you graph your data, you get the plot below. 2.5 1.5 0.5 20 40 60 80 100 120 140 160 s, met rbcoD/L (2 points) what type of kinetics are evident (I" order. 2nd order. Michaelis-Menton, Monod, etc.)? b) points) What are your best estimates of the half saturation constant maximum specific growth rate (um), and the decay rate (b), based on inspection of the above plot? (No replotting or calculations are necessary, but your answers should make sense with respect to the above plot). You can show your reasoning by marking va on the above figure. rocoolL. Explanation / Answer

a. First order kinetics is evident. The graph is obeying Michaelis-Menton's equation.

b. Half-saturation constant (Ks) will be that substrate concentration at which maximum specific growth rate is half.

Maximum specific growth rate = µmax = 2.1 mg rbCOD/L.d (should be considered slightly higher than µmax of the graph).

Ks = 25 mg rbCOD/L

(Please note: your answers are wrong).

c) Ks = S

µ = µmax [S]/ Ks +[S]

= µmax [S]/2[S]; So, µ = µmax /2

d) Yield will also be halved at Ks =S
