Question

Name the tissues that contained the highest concentration of peroxidase. Describe one function that peroxidase may perform in each of these tissues. Name one plant tissue area that contained proteins that were transferred to the nitrocellulose but did not contain peroxidase. A comparison of the relative intensities of blue spots produced by the peroxidase standards to the intensities of blue spots produced by the vegetable extracts should enable you to estimate the amount of peroxidase in the vegetable extracts. Estimate the amount of peroxidase in 1ml of your undiluted vegetable extract. Estimate the amount of peroxidase in one gram of the vegetable that was used as starting material for preparation of the extract

Explanation / Answer

1)The tissues that contained the highest concentration of peroxidase are the epidermal and phloem tissues.

2)When present in the epidermis of the plant cell wall , peroxidase helps in strengthening the cell wall by cross linking the phenolioc residues of the polysaccharides and glycoproteins components of cell wall.When present in the phloem tissues, peroxidase protects the sieve elements of the phloem from damage during oxidative stress.

3) The apical meristematic cells of the stem contained proteins that were transferred to the nitocellulose but did not contain peroxidase.

4)

A) Amount of peroxidase in 1ml of undiluted vegetable sample = (500/ t)x (1/1000) x (TV / VU )

where t = time change in minute;

TV = total volume of the extract in ml ;

VU = volume used in ml;

B) Amount of peroxidase in 1 gm og vegetable sample = g-1 f wt = (500/ t)x (1/1000) x (TV / VU) x (1/ f wt)
where t = time change in minute;

TV = total volume of the extract in ml ;

VU = volume used in ml,

f wt = weight of the fresh leaf tissue in gram.
