5. (3 points) Two proteins have been purified by a researcher, called Bucky A and Bucky B. She determines the extinction coefficient for each generating the following information Bucky A: 1 mg/ml solution OD280 1.1 Bucky B 1 mg/ml solution OD280 0.27 She then purifies a new preparation of Bucky A. Her new preparation of Bucky A has 4 ml total, and when it is diluted 1/10 it gives OD280 0.44. The total protein in her new preparation of Bucky A mg Insert the words "higher" or lower Bucky B has a fraction of Trp residues than the average proteinExplanation / Answer
The standard curve equation: y = 0.012x
where y is the absorbance and X is the protein
so x = y/0.012
= 0.44/0.012
=36.6
but it is diluted 1/10, so the total protien is 366 ug/ ml
Bucky B has a lower fraction of Trp residues that the average protein. Because, the aminoacids of aromatic rings tryptophan absorb at higher wavelength.
