Question
8. Two individuals (A and B) were infected with the HIV virus. Cells from individual A express HLA- A1l and HLA-23 proteins, while cells from the individual B have HLA-A30 and HLA-A03. The peptides from the nef protein produced by the HIV virus, which can be presented by these MHC molecules, are shown below. Based on this information, please answer the following questions. INDIVIDUAL A INDIVIDUAL B allele start end peptide IC50(nM) 1 allele start end peptide 1C50(nM 2 HLAA'11 84 92 AVDLSHFLK 15 2 HLA-A 30 19 27 RMRRAEPAA 4 3 HLA-A11 9 17 SVIGWPTVR 144 3 HLA-A03 188 196 RLAFHHVAR 80 4 HLA-A11 31 39 GAASRDLEK 1804 HLAA 30 132 140 GVRYPLTFG 82 5 HLA-A11 144 152 KLVPVEPDK 361 5 HLA A 03 84 92 AVDLSHFLK 120 6 HLA-A 23 137 145 LTFGWCYKL 409 6 HLAA03 163 171 SLLHPVSLH 147Explanation / Answer
1. HLA-A11 has a strong association with hepatitis especially autoimmune ones. It has a synergistic effect. Secondly A11 is a close part of haplotype A11-Cw4-B35-DR1-DQ1 which accounts for only second factor in rapid progression of HIV infection. HLA-A23 which is similar to A24 has a frequency to cause Kaposi sarcoma but there also it depends on more than one MHC. Whereas HLA A30 and A03 have more chances of having HIV infection.
2. As referred earlier the HLA-A11 antgen is a part of haplotype A11-Cw4-B35-DR1-DQ1 and thus the progress in HIV infection will be slower as compared to other antigen presence. Therefore, if a individual has homozygous HLA A11 there are more likely chances that the patient will remain symptom free for more than 2 years or so.
