Question

The rate of an enzymatic reaction is measured with different substrates. From the experiments, the Kw and keat values were determined for each substrate as given in Table Q.1.[c]. Kadar tindak balas berenzim diukur dengan 3 substrat berheza seperti yang diberikan dalam Jadual S.1.[cJ [c] Table Q.1.[] Jadual S.I.[c] Compound Compound Compound Sebatian Sebatian Sebatian 15 0.14 at Ku (mM) koal (s) 31 25 0.06 2.80 Based on this data, answer the following question Berdasarkan data tersebut, jawab soalar saalán berikul' i] Which compound binds tightest to the enzyme? Explanation / Answer

Please find the answers below:

Answer [i]: When a substrate binds to an enzyme, it must undergo chemical transformations so that it might get converted into a product and detach from the active site pocket of the enzyme. Hence, easier is the binding and detachment of the substrate from an enzyme, higher is its Kcat value. Thus, the substrate having lowest value of Kcat will be most tightly bound to the enzyme. Thus, it can be seen clearly from the tabulated data that the substrate A will be most tightly bound to the enzyme.

Answer [ii]: The enzyme having maximum value of Kcat will have maximum turnover number. Thus, it can be seen from the tabulated data that substrate C has the maximum value of Kcat, hence would be most efficient substrate for this enzyme.
