Ans1. 1. pGLO plasmid - Bacteria will grow easily on a plate with ampicillin as the pGLO has insert in the Multiple clonng site that inactivate the ampicillin resistant gene or marker gene. This will lead to the growth of bacteria on LB plate containing ampicillin.
2. Bacteria will grow on the second plate too as there is ampicillin and arabinose in the media and pGLO is also having the gene responsible for the growth in presence of substrate arabinose and ampicillin marker gene.
3. Bacteria will not grow on this plate as there is no kanamycin marker gene in the pGLO, so the bacteria will be susceptible for kanamycin and hence will die and no growth will be seen on LB.
Ans 2. 1. Grown bacteria are the transformed, so it will glow green due to presence of GFP reporter gene.
2. Again there will be transformed bacteria as only they will grow in this plate and will glow.
3. No growth and glow will be seen.
