Cell & Molec Bio Lab
Using the data from the table in the picture, how many different dyes were resolved on the reversed phase minicolumn? What were their colors?
NAMES: PART 3. (Use this page as part of your lab report) NAME OF "UNKNOWN" DRINK MIX: Read the absorbances of cach of your fractions using the wavelengths indicated in the chart below. You might need to dilute them with some water to get enough volume (2 ml) to read in the spec. Some samples might give very high readings. If so, dilute them and try again. Vortex well to mix in cither case. You will probably have fewer fractions than those listed, maybe just 4 or 5. Don't worry about this. You sce what you see in this, as in everything else!! Please refer to the chart below. 61. "Flow through" means the liquid that came off the column when you put the diluted dye mix through it. This contains any dyes that "flowed through" the column without sticking to it. "Fraction" is just a way to refer to the tube number, with fraction 1 containing the material that came out ("eluted") first, etc "Elute" means that the molecule comes off the column."Eluate" is the material that cluted (came off).Explanation / Answer
From the picture it is clear that the reversed phase minicoloumn is the flow through column. Four different colors are evolved in dying. The colors Magneta, Green, Cyan and Blue. Why because when red and green mixed, yellow ellutes.,red and blue ellutes magneta and finally blue-green combine to give Cyan.
