Question
B1B1 B1B2 B2B2 [Enzyme] An experiment was conducted to extract the "Bee" enzyme from bees and measure their activities. All experiments were conducted at 25°C. Extracts were obtained from bees with different genotypes (different combinations of alleles) and diluted to different concentrations. Enzyme concentration (represented on the x axis) has been controlled for in this assay: therefore, on each point on the x axis, all three samples contain the same amount of enzyme. In this bee population there are two different alleles: B1 wild-type and B2 mutant. (total 3 marks) A. Based on the results shown in the figure, what type of mutation is most likely responsible for this observation? (gain-of-function, loss-of-function, temperature-sensitive mutation, overexpression, etc). (1 pt). Explain your choice (1 marks) B. In separate genetic experiments, a third allele (B3) was discovered. The DNA sequence of this allele reveals a mutation in the promoter region of this allele Will the product of this allele have a similar or different activity profile as gene products of other alleles of this gene? Show your answer by predicting the activity profile of this enzyme (as a function of enzyme concentration) directly on the graph above and labeling it, if the enzyme extract was obtained from a bee with a genotype of B3B3. (1 marks)Explanation / Answer
A. Loss of function mutaiton
All experiments were conducted on same temperature, thus loss of function is not possible dependenet on temperature.
B1 allele is wild type, thus B1B1 allele shows wild type enzymatic activity.
B1B2 is heterozygous, and shows reduced enzymatic activity whereas
B2B2 showd mutant enzyme activity, which showed loss of function activitity compared to wild type.
B.If B3 allele has a mutation in promotor sequence, it can cause a functional change in the enzyme activity frofile, reducing the activity of the enzyme. Thus , bee with genotype B3B3 might showed similar genetic activity like B2B2. The grapg for B3B3 will be similar like B2B2.
