Question
Write 2-20, 20-200 ., or 100-1,000 . to indicate which micropipe should be used for each of the following measurements: 0.015 ml2 20ul 1.0 mL-100-106( 1 b tech working at a medical lab, who did not pay close attention during her cellular and molecular biology class in college, has been using the micropipettors incorrectly. h down to the second stop both when getting ready to fill the tip and when emptying the tip. Design and an experiment to determine how her mistake affects the accuracy and precision of her measurements. 4. A la 14Explanation / Answer
According to the given problem, the student is using micropipette wrong, and she pushes button to the second stop during filling and emptying the pipette.
Doing so will definitely let her fill more volume than the set value on the micropipette, leading to the incorrect measurement and consequently false results.
For example,
She has to prepare a buffer of 1X concentration with volume 1ml from 50X concentration stock solution.
Then for this she has to use formula
M1V1 = M2V2
1*1 = 50*V2
V2 = 2/50 = 0.02ml
So she has to take 20 microlitres of stock volume and then need to make up the volume upto 1ml by adding 980 microlitres of the diluent.
So she will use a pipette of 2-20microlitres and the other of 100-1000 microlitre.
But she is pressing it to the second press, so she will take more volume than the 20 microlitre as well as frm 980 microlitre.
Due to change in the volume she will prepare a working solution different from 1X. This will cause deviation in the standard experiment.
