There are many enzyme assays can be performed in the laboratory by which we can analyze enzyme activity. These assays are coupled with spectrometry, fluorometry, calorimetric etc. Enzyme activity is measure by the given formula
The mole of substrate converted per unit time= rate x reaction volume
Some standardized substrates are analyzed for the enzyme activity such as gelatin and milk protein.
There are six classes of enzyme which are as follows-
1. Oxidoreductase
2. Transferases
3. Hydrolases
4. Lyases
5. Isomerases
6. Ligases
Gene expression can be analyzed by many techniques such as RT-PCR, northern blotting, qRT-PCR, FISH, RNA- seq etc.
Northern blotting- In the process of northern blotting the study of gene expression by detecting RNA can be done. The process of northern blotting includes the use of electrophoresis to separate RNA fragments on the basis of size of the RNA fragments and the detection is based on the hybridization probe.
RT-PCR- Reverse transcription polymerase chain reaction (RT-PCR) is a susceptible assay for the rapid, economical and easy detection of gene expression by RNA. It has ability to amplify numerous templates in a single reaction cycle.
Reverse transcription polymerase chain reaction (RT-PCR), an alternative of PCR which is used in molecular biology to detect RNA expression. There is a quite different technique called as qRT-PCR. The RT-PCR is a quantitative detection method in which the gene expression is analyzed by using cDNA whereas in the qPCR, the quantitative measure of the amplification of DNA can be done by using fluorescent dyes.
FISH- Fluorescent in situ hybridization (FISH) is based on the hybridization with the specific sequences by the use of fluorescent probes. It is used to identify and localize the occurrence or nonexistence of specific target sequences. After the hybridization with the specific sequences by using fluorescent probes the binding of the fluorescent probes can be analyzed by using fluorescence microscopy.
RNA- sequencing (RNA-Seq )- It is also known as whole transcriptome shotgun sequencing (WTSS), which is used to disclose the attendance and amount of RNA in a given sample. RNA-Seq can also be analyzed the constantly changes which occurs in cellular RNA expression. This technique can also be used for the analysis of the introns and exons.
