Question 1 Describe an experiment that you could perform to answer the questions po posed below. You may use microscopy.phase-contrast micros.copy, Bifferential interference-contrast microscopy ned bright field microscopy. fluorescence microscopy (epi microscopy (transmission or scanning), XRay Crystallography, or Differential your reasoning behind your choice. a. Is amino acid 6 (glutamic acid) in human hemoglobin part of an alpha helix or a -sheet? b. Is Smad1 (protein) located in the nucleus or the cytoplasm of cultured fibroblast cells? c. Does OPA1 mutation affect the number of cristae (membrane infoldings) in mouse liver mitochondria? Does a mutation in the fly gene sevenless affect the hexagonal 3-dimensional geometry of the fly eye cells? d. e. Does mutation in N-acetylglucosamine-1-phosphotransferase (mutated in the human disease mucolipidosis type I) affect the density of lysosomes? f. Is a newly discovered single-celled organism a prokaryote or eukaryote? g. Do cancer cells migrate faster than non-cancer cells?

Explanation / Answer

a. the 6th amino acid (glutamic acid of beta chain) is a part of alpha helix.

b.Smad1 protein is found in both cytosol and nucleus during resting stage. But during signalling (by bone morphogenetic proteins, BMP) they get concentrated in nucleus.

c. Optic atrophy 1 (OPA1) is the mitochondrial GTPase. It is found in the inner membrane of mitochondria and is resposible for mitochondria fusion. Mutation in OPA1 causes changes in the structure of cristae and accelerates the release of cytochrme c upon the induction of apoptosis.

d. The sevenless gene in D. melanogaster encodes for receptor tyrosine kinase protein. It is essectial for the formation R7 cells in the eye. Loss of function mutation in seveness gene results in failure to differentiate photoreceptor cell called R7.
