a) What kind of inhibitor is P56?

(b) Use the equations of the lines provided to calculate KM and V max for the enzyme with and without the inhibitor.

(c) Calculate the value of KI in the presence of 0.22 mM inhibitor.

please answer fully a-c. thank you

54. A cysteine protease from a malaria parasite was recently iso- lated. A compound named P56 was found to inhibit the enzyme. Compounds of this type have the potential to be used as drugs to treat malaria. (b) Use the equations of the lines provided to calculate KM and Vmax for the enzyme with and without the inhibitor (c) Calculate the value of K in the presence of 0.22 mMM inhibitor

Explanation / Answer

Ans a) P 56 was found to inhibit parasite of malaria cysteine protease and it have the potential to be used as an antimalarial drug.the mechanism of action of p56 as dock protein downstream of receptor or non-receptor tyrosine kinase.

So p56 act as a competitive inhibitor because as show in figure Km is increased and the Vmax remain the same.

Ans b)
for Inhibitor P56-
the KM = -(1 / -0.5µM-1) = 2 mg/ml
and Vmax = 1 / 2.35 mM-1*s = 0.425 µmol/µl

for No inhibitor-
the KM = -( 1 / -1.2µM – 1 ) = 0.83 mg/ml
and Vmax = 1 / 2.25 mM-1*s = 0.44 µmol/µl

Ans c) Calculation for the value of Ki in the 0.22 mM inhibitor presence .

becoz 2 mg/mL / 0.83 mg/ml= 2.40 = alpha

and Ki = 0.22 mM / 2.40 -1 = 0.1571 mM
