Question

You find a mutant single-celled amoeba that does not move in response to addition of a food source. With further experiments, you figure out that there is a defect in one of the membrane-tethered Rho family proteins specifically related to its ability to interact with the GEF.

(A) What are the three Rho family proteins? How would a mutation in each of them inhibit the amoeba’s ability to migrate towards the food source?

(B) What effect would addition of nonhydrolyzable GTP have on a normal amoeba or on this mutant amoeba?

Explanation / Answer

a.) Rho family proteins are small G-proteins. Three main rho proteins are Cdc 42, Rac1, Rho A. Mutation in these protein will disturb the movement of Amoeba by altering the movement of actins.

b.) Rho family protein are small GTPases are unable to cause signaling cascade in unhydrolyzable GTP as these cant phosphorylate the Rho protein in both normal and mutant.
