Your lab partner accidently set the spectrophotometer to measure % transmittance instead of absorbance, and all the numbers you recorded were % transmittance, what kind of trend will you see in your data? Does this affect your overall conclusion of the experiment (causes of cell membrane 3. damage)? Explain 4. If not all of the betacyanin were properly rinsed off the beet cylinders, how might your results be altered for the temperature-treated investigation? How might your results be altered for the solvent-treated investigation? Explain your reasoning.

Ans 3. spectrophotometer is a device to calculate the concentration of any substance. To calculate the concentration the device use the principle of absorption of light by the sample. every substance absorbs maximum light of a particular wavelength only, which is fixed for a substance.So amount of light absorbed is directly proportional to the concentration. absorption formula is negative concentration of transmittance(amount of light given out by a substance).So if we calculate percentage transmittance then the we will see low values on spectrophotmeter for all the data.This can affect the conclusion since the concentration of the substance is affected.

4. The beet is cut into cylinders because of cutting betacyanin will be released which should be washed out completely.If betacyanin is not rinsed out then it will interfere in the reading of spectrophotometer.The reading will be more than what should be observed in both cases temperature treated and solvent treated.

7.Yes there is relation between the degree of damage and temperature.It will be observed that if we increase the temperature then colour of betacyanin in the test tube at respective temperature will be more dark so more light will be absorbed.so less light will be tranmitted.This happens because more cell damage increases with temperature and more betacyanin will be coming out so more absorption will occur.

8. According to me more heat will cause more damage to cell membrane.low temperature can cause stiffness but after the cell comes to normal temprature the cell will regain its fluidity but more heat will permanently damage and denature the proteins.

9. All membrane can tolerate temperature at certain extent so there can be temperature which will be safe to the membranes.

10 Heat can affect the proteins of the membranes.The membrane are made up of proteins which are sensitive to heat.As heat increases the proteins starts to denature and thi can cause death of the cell.
