Lab Challenge 2A&B; (10 pts total) Name: You will be given maps for four plasmids and a plasmid DNA sample that is one of the four plasmids for which you were given maps. For Part A, you are expected to design a set of experiments that will enable you to identify which of the four plasmids you were given. For Part B, you should come up with a list of reagents you will need to carry out your experiments. A maximum of 10 points will be awarded for designs that would be effective. Over the following two days you will be given time to execute your experiments and come up with data that you will analyze to determine which plasmid you were given. Successful identification of the plasmid (based on the data you supply with your answer - successful guesses don't count!) will earn you the remaining 30 points.Explanation / Answer
Step 1 : Select one restriction enzyme which has restriction site in all four plasmids.
Step 2 : Count the number of restriction site that a selected enzyme has in each plasmids. For example, if the selected restriction enzyme has only one site in a particular plasmid then it must result in only one fragment when digested with that enzyme.
Step 3 : Carry out restriction digestion for all four plasmids and to your given DNA sample.
Step 4 : Run the gel electrophoresis technique with all five samples.
Step 5 : Compare the results or number of fragments obtained from given DNA sample to other four plasmids.
