You are studying some Martian cells with similar chromatin and DNA to life found on Earth. You set up a "Mars growth chamber" to grow your cells at 10 degrees C (they like it cold!!).You are extremely interested in the ALN gene.
a) You find ALN is not being actively transcribed in your cells, however, when you perform chromatin immunoprecipitation, you find that RNA polymerase is bound to the ALN promoter. Explain these results.
b) Oh no! A summer high school student has taken your Martian cells and moved them into the 37 degree C incubator! Given you observations above, what change would you expect to see if you monitored the ALN gene in these cells by quantitative PCR? Please expain as clearly as possible. Thanks!

1. ALN gene not transcribed because some inhibitor molecules inhibit the ALN promoter or transcription factor not activated inside the cell. We need to add the necessary growth factor to activate the pathway to release the inhibitor from the promoter region or to release the transcription factor from the inhibitor to activate the promoter.
2. Martian cells with similar chromatin and DNA to life found on Earth so it can able to grown on wide range of temperature or slows the growth rate of the cells. PCR we have doing 3 stages, Denaturation, annealing and extension.
In Denaturation we have to use high temperature upto 950C . It is similar to our DNA so there is no problem in DNA synthesis in PCR. There is increase in copies of ALN gene in quantitative PCR.
