Write (or type) your nswers on a separate piece of paper. Each student must write their own answersl Fill in the Table below. Sclentists)Experiment (general description) Archibald Garrod Conclusions Beadle & Tatum Srb & Horowitz 2. What is meant by transcription and translation and how are these two words related to Mende's genotype and phenotype concept fi.e. how might a change in the genotype lead to a change in phenotypej?
Explanation / Answer
1.archibald garrod experiment-Garrod is best known for study of inborn errors of metabolism. He developed an increasing interest in chemical pathology, and investigated urine chemistry as a reflection of systemic metabolism and disease. This research, combined with the new understanding of Mendelian inheritance, evolved from an investigation of a few families with an obscure and not very dangerous disease (alkaptonuria) to the realization that a whole territory of mysterious diseases might be understood as inherited disorders of metabolism.
conclusion-he developed possible nature of inherited diseases of metabolism. He described the nature of recessive inheritance of most enzyme defects. Garrod expanded his metabolic studies to cover cystinuria, pentosuria, and albinism. These three inborn errors, along with alkaptonuria are collectively called Garrod's tetrad.
2.beadle and tatum experiment- genes act through the production of enzymes, with each gene responsible for producing a single enzyme that in turn affects a single step in a metabolic pathway.this is called one gene one genome hypothesis.
conclusion-Presently, the one gene–one polypeptide perspective cannot account for the various spliced versions in many eukaryote organisms which use a spliceosome to individually prepare a RNA transcript depending on the various inter- and intra-cellular environmental signals.
3.srb and horowitz experiment- ound that Arginine is synthisized in a metabolic pathway that involves ornithine and citrulline as intermediates
-Grew mutants in media containing or lacking either ornithine, citrullinem or arginine
conclusion - None of the mutants grew on minimal medium alone but, mutants grew on media supplemented with either ornithine, citrulline, or arginine arg 2 mutants grew on media supplemented with either citrulline or arginine arg 3 mtants grew on media supplemented with arginine
Proved one-gene, one-enzyme hypothesis.
