Question

In lecture we were discussing a sample problem where several of you had infections with retroviruses. I believe that for that example, Rachel had a case of hepatitis C. Hailey came in and we knew she had a virus and thought that it might be hepatitis C based on other tests performed. Yan performed an in situ in order to determine whether it was the hepatitis C virus. She used Rachel's tissue as a control sample. The virus was detected using a probe that binds all viruses and shows up as red, the cells of the tissue will be detected using yellow, and the blood vessel cells will be detected using blue. (red and yellow makes orange, red and blue makes purple) a. (2 pts) What tissue will Yan be pulling for the biopsy? ___________ b. (2 pts) Will she be testing for DNA, RNA, or protein and give some explanation for this. c. (2 pts) Based on Rachel's results, where at in the tissue should the hepatitis virus be found? d. (2 pts) Based on Hailey's results, does she have hepatitis C and why? e. (2 pts) Based on Hailey's results, do you think she only has a viral infection in this organ and why?

Explanation / Answer

1. A liver biopsy is used to diagnose cirrhosis, which is a condition where liver cells are destroyed and renewed by scar tissue because of chronic inflammation. The scar tissue alters the flow of blood and other fluids through the liver. Without good blood flow reducing in total liver cells, the liver hence cannot function properly and becomes lumpy and hard. Scar tissue can form in the liver due to inflammation that happens beside hepatitis C.

2. The presence of HCV RNA is needed to prove chronic HCV infection.

3. HCV RNA could be detected in the serum of patients who were positive in liver tissue.

4 The probe assay includes ferrocene-labelled signal probes to detect the six major genotypes and their most common subtypes. Haley does not have hepatitis c as other tests showed the negative result as the probe did not change its red colour to purple.

5. liver inflammation can be a cause of viral infection by other hepatitis or different viruses in the liver and since she has the virus in her system it is a viral infection but not hepatitis c.
