Question
Which structural state(s) is expected for each specified enzyme entity, under each of the following conditions. (a) ATP synthase beta subunit as found in mitochondrial membrane; (b) ATP synthase beta subunit in a6b6 hexamer with no gamma subunit; (c) ATP synthase alpha subunit; (d) c subunit in ATCase holoenzyme under low [L-Asp]; (e) c subunit in ATCase holoenzyme under high [ATP] and high [L-Asp]; (f) c subunit in c3 trimer separated from other proteins. (g) star the states that are high-performing from a kinetic point of view.
Explanation / Answer
(a)ATP synthase found in the mitochondrial membrane and consist of two components -Fo and F1 ATPase.Fo is embedded in membrane while F1 is protrude towards matrix .Fo consist of one a and two b ,9-12c subunits.F1 having 3alpha ,3beta ,gamma ,delta and epsilon.Beta subunits are the sites of ATP synthesis.
(b) a6b6 hexamer is a holoenzyme of GCC(geranyl-coA carboxylase) which is first purified from the Pseudomonas .It consist of two, alpha and beta subunit and holoenzyme as a6b6.Beta subunit contain N and C domains of carboxyltransferase.
(c)ATP synthase having two alpha helices present in the c subunit ,which is present in the mitochondrial membrane. There are 9-12 c subunits .Gamma epsilon and C subunit act as rotor or moving unit.
(d)ATCase or aspartate transcarbamolyase enzyme involved in pyrimidine biosynthesis.it is made of six regulatory and six catalytic subunit present in dimer of 3 .It is present in two state, T (tight) and R for relaxed state.T state is characterized by low substrate affinity and low catalytic activity and R in opposite manner.Under low (L-Asp),enzyme is present in the T state and then converted into R state.
