This medium is prepared as basic to determines the oxygen requirement of different bacteria ( both gram positive and gram negative bacteria).The contents of the medium are : thioglycollate and L- cystein that make the anaerobic environment , diminishes the oxygen by converting the oxygen into water. Agar is also used in the medium in small amount and the Resazurin as the oxygen indicator. Pink colour indicates the presence of oxygen.

Tube 1 indicates that all organism grow at the top where oxygen is most ; means they are obligate aerobes need oxygen to survive.

Tube 2 shows obligate anaerobe ; will be seen at bottom where oxygen is almost not found or in lowest concentration.

Tube 3 show the facultative anaerobes : they grow either in presence of oxygen or may in absence of oxygen. But if oxygen is available they respire aerobically .They gather at the top in tube because there is higher production of ATP in aerobic respiration than anaerobic.

Tube 4 shows the microaerophiles .They neend oxygen but only in considerable amount. Higher amount of oxygen is poison for microaerophilles. They found between middle and top of the tube according to their need of oxygen.

Tube 5 shows aerotolerant anaerobes : They live uniformly throughout the medium, not affected by presence of oxygen and also not require oxygen.
