In case of top bands, as indicated its unprocessed VSV-G sample, top band of western blot represented the free heavy chain molecule of the antibodies, which are higher molecular weight protein molecule that were abundant (unbound stage) at the initial. Later as the incubation proceeded, the immunoprecipitation was significant due to antibody binding to the target protein and precipitation was effective resulting in the increased amount of the immunoprecipitation for the target protein. Therefore, the bandwidth of target protein presented in the western blot analysis was increased across time of precipitation as the time interval increases.
