History Bookmarks People Window Help meheleDownloads/Chapter%2 2018%20(2).pdf Q: Why are eukaryotic transcripts more stable than prokaryotic transcripts? A: Q: What type of protein would you expect to have greater stability, a primary protein or one with a quaternary structure? A: Q: Jakob-Creutzfeldt Syndrome is the human manifestation of bovine spongiform encephalopathy, a protein-folding disorder. What type of protein-based medicine might prove beneficial for treating the disorder if one could figure out how to target the prions?

Explanation / Answer

Answer 1. This is dependent on the gene expression regulation pattern of both the types of organisms. Prokaryotic gene expression is mainly regulated in transcriptional level. This is possible if mRNA half life is short. Only when an RNA is unstable its accumulation depends on transcription.

On the other hand Eukaryotic gene expression is regulated on translation level, which is only possible when mRNA is stable. Stable mRNAs can be activated/deactivated by translation.

Answer 2. A protein with quaternary structure will be more stable because a protein in quaternary structure will be stabilized by various stabilizing forces due to bonding interactions between the side-chain groups of the amino acids. These forces include hydrogen-bonding, disulfide-bridges, vander waal forces and salt bridges. However a primary structure protein will be a linear chain protein and will not be stablized by these interactions and thus will be prone to degradation.

Answer 3. Creutzfeldt-Jakob Disease (CJD) gradually destroys brain cells.  It is caused by an infectious agent, called a prion. There is no cure for CJD yet and only option is pallative care.

A protein based medicine that degrades the prion protein named Prion Protein (PrP) may prove beneficial in treating the disease. Other protein based medicines that block the interaction of PrP with chaperons can also prove beneficial to treat this disease because prions exploit and interact with chaperons to misfold the healthy proteins in the host.
