04. Prepare Results and Discussion Section based on provided figures and information Guide is provided on the next page. . You will have to summarize the figures based on directions · Include sentences in yellow and respond as requested Results and Discussion The PIF4 protein regulates Arabidopsis thaliana hypocotyl and petiole growth, especially at elevated temperatures. From a genetic screen of PIF4 overexpression background lines containing T-DNA insertions, we recovered several mutants that displayed altered plant growth or altered PIF4 protein accumulation. One mutant was deficient for the protein PWR (powerdress) and displayed suppressed growth at 23'C and 27°C (Figure 1A and 1B). Comment on mutants compared to Col-0 for hypocotyl and petiole growth. It has previously been reported that PWR interacts with the protein HDA9 (histone deacetylase 9). Thus, interactions between PWR and HDA9 are likely required for PIF4 medidated-plant growth as revealed when Col-0 and hda9-1 were grown at elevated temperature (Figure 1C). Provide a working model addressing PWR protein recruitment of HDA9 proteins to suppress transcription as it relates to euchromatin regions and heterochromatin regions. We also recovered two mutants in which PIF4 protein expression was altered (Figure 2). These mutants are either deficient in the protein DET1 (De-Etiolated 1) or the protein COP1 (Constitutive Photomorphogenesis 1) Comment on PIF4-HA protein expression in det1-1 35S PIF4-HA plants and cop-4 35S PIF4-HA plants compared to 35S:PIF4-HA plants. Comment on DET1 protein and COP1 protein as it relates to the expression or accumulation of PIF4 protein. Provide a working model addressing DET1 proteins and COP1 proteins recruitment of regulatory proteins for gene expression or protein accumulation.

Explanation / Answer

PIF4 gene is involved in the growth of plant by developing the petiole and hypocotyls of plants. These genes works best with the increase in temperature and more temperature favors the function of these genes. Also for the expression of PIF4 gens the interaction of PWR and HDA9 gens are essential any mutant in the genes of PWR and HAD9 will lead to the low expression of the PIF4 leading to less hypocotyl growth which can be seen from the graph in Fig 1A were PWR gene was mutated which affected the PIF4 expression leading to less hypocotyls length even under high temperature or low temperature. Same like PWR gene, expression of Col-0 gene is also required for the expression of the pIF4 gene under the gene expression of the Col-0 a significant of hypocotyls length is seen. These genes also affected the petiole the wild type Col-0 showed normal petiole growth under high temperature while the strains with mutated genes in PWR and pif4 showed less petiole growth. The mutant strain with mutated gene Hda9 also showed less growth in high temperature.

The DET1 and COP 1 works under dark conditions favors the growth in dark conditions while the with favorable light the PIF4 expression is seen in greater amount and more amount of protein accumulated with more gene expression under both temperatures of 220C and 270C in SDS PAGE.
