Discussion:

Catalase: The catalase test is used to distinguish between those micro-organism that produces the enzyme and are able to break down the hydrogen peroxide into oxygen and water. Both Micrococcus luteus and Enterococcus feacalis are catalase positive, and this means that the culture did not contain any of the micro-organisms.

Oxidase: The oxidase test determines the presence of various cytochrome forms, and from the experiment, it was evident that both E. coli and P.aeroginosa are oxidase positive.

IMVic test: The test incorporates various sub-tests including indole, methyl red, Voges- Poskauer, and citrate. The test can be used to differentiate between E. coli and P. aeroginosa. E.coli tests are positive for methyl red and Voges- Poskauer tests. On the other hand, P. aeroginosa test positive for the citrate test.

Conclusion:

Based on the test results, the most identified micro-organism was E. coli. The serial testing is done to isolate and rule out the most suspected pathogens in a cultured colony. The micro-organism tests positive for the most of the IVMIC tests as evident in the above summary. The biochemical tests are based on the ability of the bacteria to produce given chemicals that takes part in the reaction; the chemicals may be enzymes which initiate catalysis or breakdown of the substrates provided in the test reagents. E. coli is a common Enterobacteriaceae and commonly available on the body surfaces and water. The other tests are based on the gas production as a result of the metabolism process which results to the change of the culture media into either and acid or an alkaline.

Explanation / Answer

To distinghuish whether the microorgansm has catalaze enzyme or not, a test known as catalaze test is performed. This enzyme breaks the hydrogen peroxide into oxygen and water.Since Micrococcus luteus and Enterococcus feacalis are catalase positive, the sample that was tested for catalze positive microorganism, did show any catalase positive bacteria.

similarly, to test another enzyme oxidase, oxidase test was performed.  E. coli and P.aeroginosa were found to be oxidase positive.

MVic Test: This test is an assembly of many other tests which differentiate between E.coli and P. aeroginosa. Among the various tests, E.coli is positive for  methyl red and Voges- Poskauer tests whereas P. aeroginosa test positive for the citrate test.

Conclusion:

The most identified bacteria was E.coli. E.coli is a common bacteria that is present both in the internal environment and outer surface of the body area. Various tests were employed to decipher the colony of the bacteria. Some of these tests were based on the production of the chemical that was produced by the enzyme present in particular species whereas some were based on gas production as result of metabolic processes eventually changing the pH of the culture media.
