A digital ELISA experiment was used to quantify the concentration of antigen in a 100 uL sample prepared with 10,000 magnetic beads decorated with capture antibodies, excess detection antibodies with biotin tags, and excess streptavidin tagged enzymes (analogous to Figure 3 in the reading). After rinsing the magnetic beads, 100 were selected and each sealed into an individual well with enzyme substrates that yield fluorescent products.
