Question

80 (6pts) In this figure they are examining 3 potential binding sites for ??-i (R87) (K321) (K447) The three mutants each correspond to the mutations shown in A, and represent the 3 potential binding sites (i, ii and i). They do a Co-IP of the Ciz1 protein and its mutants, and look at the levels of cyclins that are recovered. Which binding sites on Ciz 1 are important for Cyclin E binding? Which are important for Cyclin A binding? How did you know? Mutant Cyclin E Cyclin E Mutant Cyclin A Cyclin A You hypothesize that Ciz 1 binding to cyclin A changes the cyclin/CDK localization allowing for proper cyclin/CDK function. Using any of the tools that are shown above, and based on your previous answers design an experiment to test this hypothesis. Describe your experimental design (4pts), Draw and label the figure that would result from this experiment should your hypothesis be true (4pts), Write a figure legend to accompany that figure (4pts) h.

Explanation / Answer

The mutant Cy ii shows no or less recovered cyclin E protein on the gel. Which means that there is no cyclin E binding due to mutation in Cy ii region of Ciz 1. On the other hand, other mutants cy i and cy iii showed recovered protein even after mutation. Hence the cy ii region in Ciz 1 is important cyclin E binding.

Cy iii region is required for cyclin A binding. As the mutant Cy iii shows no or less recovered cyclin A protein on the gel. Which means that there is no cyclin A binding due to the mutation in Cy iii region of Ciz 1. On the other hand, other mutants cy i and cy ii showed recovered protein even after mutation. Hence the cy iii region in Ciz 1 is important cyclin A binding.
