Naffie Section: Date BMS 504 General Microbiology Laboratory Laboratory 13 Questions - Water microbiology 1. Why are laboratory tests for bacterial pollution of waters directed to the detection of coliform bacteria? And why is Escherichia coli used as an indicator organism in microbial analyses of water? 2. What procedures are used to ensure the safety of potable water supplies? 3. What implications would the presence of coliforms have in human food and water consumption?

Explanation / Answer

1. Colliform are large group of bacteria that are found mostly in sewage and animal waste. In addition to the colliform, sewage water also contain other harmful bacteria too. When there is a presence of colliform in water, it indicate that other harmful bacteria are present in the water. Hence, whenever in the drinking water, there is a presence of colliform bacteria it show the presence of impurity in the water. Bacterial colliform impurity can be checked by calorimetric test. Change in the color in calorimetric test show the presence of colliform bacteria and thus the impurity in water.

E. Coli presence is the indicator of contaminated water. E. coli is a member of colliform fecal bacteria, hence presence of E. coli indicate the presence of impurity in drinking water as compared to the presence of other bacteria.

2. Protection of water from contamination, removal of contamination with the help of various treatment methods, presentation of water during water storage, distribution and handling of water are important procedures used in the treatment of water. Other procedure include abstraction, treatment and distribution of water. Identification of potential sources of protection is also one of the method.

3. Implication that presence of Caliform bacteria in water and food indicate that both are contaminated with many impurities and not good for consumption as it can cause many diseases like food poisoning. Colliform are large group of bacteria present in sewage and animal wastes. Whenever, colliform bacteria is there in food and water, it also show the presence of harmful bacteria. Hence, these type of contaminated food and water should be avoided.
