To study this gene’s expression during embryonic development, would you choose to fuse a reporter gene, like GFP, with the promoter region or the enhancer of your gene of interest? Reporter gene like GFP should be fused with promoter regions. Because RNA Pol binds with promoter and start processing downstream from there, so if we put GFP in the downstream of promoter we will be able to study the gene expression.as enhancer region is further upstream of promoter so GFP fusing in this area will not work.
