Question
t-0 t 20 sec 26. You have carried out a polarity-marked microtubule gliding experiment as we discussed in class with conventional kinesin (Kinesin I) and a mutated form of Kinesin I that you happen to be studying. Shown to the right for the two experiments are still images from the time-lapse movies. (6 pts) (1) Which experiment, A of B, was carried out with conventional kinesin 1? (2) How can you tell? (answer below) (3) For the other experiment in which the mutant kinesin was used: Which domain in Kinesin I do you think was mutated to cause the observed result?Explanation / Answer
1) Experiment A
2) Kinesin I move towards the positive end along the microtubule carrying the organelle and in the experiment A the kinesin I is shown movement along he microtubule with positive end.
3) In other experiment B the effect of mutant kinesin is seen where the movement of kinesin required the movement of neck of the kinesin proteins which allows the movement of kinesin protein in the positive direction but in the mutant due to defective neck region the mutant is seen in the reverse direction.
