Question
How do you arrive at this answer?
"GO-Slim" is a reduced subset of the Gene Ontology that is used for very basic annotation. As of 2017 it contains only 150 terms.
Suppose I have identified a set of transcripts that are differentially regulated in an experiment that I performed. I want to see if these transcripts are enriched for any of the 150 terms in GO-Slim.
I have a frequentist significance test for statistical enrichment that I can perform on each of the 150 terms, and I can choose the significance threshold (p-value) to use for this test. In choosing this threshold I apply the Sidak correction to correct for the fact that I am repeating the test 150 times.
If I want the overall p-value for all of GO-Slim to be 0.05, what p-value should I use as a significance threshold when testing each individual term? Give your answer to one significant figure.
ANSWER: 0.0003
Explanation / Answer
let significance level for each indiviudal =p
therefore type I error =probability that at least one subset will get rejected when it should not=0.05
1-P(none will get rejected)=0.05
1-(1-p)150 =0.05
(1-p)150 =0.95
(1-p)=(0.95)1/150
1-p=0.9997
p=1-0.9997=0.0003
