Question
Hi,I'm trying to understand a research paper that talks zebrafishand creating MZ Dicer mutants.
The paper says that wild-type zebrafish embryos depleted oftheir germ cells served as hosts for germ cells from homozygousdicer mutant donor embryos.
With that said, a question asks: "In order to generate the MZDicer mutants, the authors crossed fish that had a wild-type somaand a homozygous Dicer mutant germ-line. TRUE OR FALSE?"
I am thinking that the answer is false because doesn'twild-type soma indicate that the embryos contain their germ cells? And since the paper says that the wild-type embryos were depletedof their germ cells, doesn't that go against wild-type soma?
Please help.
THANKS!

Explanation / Answer
I think what the question is getting at is that typically wild-typezebrafish had their naturally wild-type germ cells removed andreplaced by mutant embryos (homozygous germ cells). That being said, I would say that the answer is TRUE, because it istrying to establish the fact that reproduction depends only on thegerm cells, regardless of somatic cells. In this case, thewild-type fish with mutant germ cells would cross, and since theyare homozygous for the mutation (aa x aa), assuming this is asimple cross, the offspring would indeed be Dicer mutants.
