The principle is based upon counting particles which are measured by changes in electrical impedence produced by non conductive particle. The non conducting particles are suspended in an electrolyte. The change in electrical impedance is directly proportional to the volume of the particle travelling the orifice. The elctrolyte solutions serves as the passage for easy flow of the particles and help electricity to flow through them; hence changes in electrical impedence are easily measured. Also, cells without isotonic solutions serve as poor conductors which alter the cross section of microchannel. Moreover, the resistance increases leading to a decrease in current flow.
