B. The positive results are obtained from posotive control. They could be from contamination, over washing, under washing etc. A positive result from posotive control indicates the procedure is optimised and is in a good working condition.

C. The ingredients in negative control wells was found be known concentrations of target proteins like murine .

D. Negative control doesnot express the protein you are detecting. It is used to check non specific binding and false positive results. Each plate you use for the experiment should contain negative control sample in order to validate the final results.
