2. The sequence has exons along with a cap and AAAAA tail. In bacteria, post transcripptional changes wont occur. So, capping and poly AAAAA tail wont be added naturally. That is why we need to put this in our construct. Splicing also won't happen in bacteria, so we choose already spliced construct.

4. genetic code is same in humans and bacteria. So, the insert will produce the same amino acid sequence as of human. But, there won't be post translational modifications. If the secreted protein will be active or not we are not sure and acquiring right conformation also depends on post tranlational modifications whihc won't take place in bacteria.
