Question

To test the role of glycogen phosphorlase during exercise, you generate a mutant strain of mice that has very low levels of active glycogen phosporylase enzyme. These mice are subjected to intense, anaerobic exercise for varying periods of time. During this exercise period, blood is withdrawn every few minutes andtested for lactate. Samples from mutant mice are compared with control samples from wild-type mice (that have normal glycogen Phosphorlase). The results are shown in the figure to the right. Why does the blood latate concentration increase brieflyin the control (wildtype) mouse Be specific as to the source of the lactate and the pathway(s) involved int he production of lactate. Why is, there no corresponding increase in blood lactate concentration in the mutant mouse

Explanation / Answer

in the wild type mouse , there was increased concentration of lactate present , because in the wild type there was normal glycogen phosphorylase which was able to peerform glycogenolysiis ( breakdown of glycogen into free glucose ) which became the source of lactate , glucose ------ pyruvate which in the absence of oxygen got converted into lactate. therfore in wild type there was high concentartion of lactate was found.

in mutant type , there was absence of glycogen phosphorylase , which was not able to break glycogen into glucose which was further not able to convert into pyruvate , which was not able to convert into lactate..therfore lactate was not found in the mutant type.
